DUALITYBIO-B (09606) has established a global research and development collaboration and licensing agreement with Genentech to develop the next-generation ADC based on the company's new DUPAC payload platform.
Ying'en Biotechnology-B (09606) announced that on August 27, 2026, the company entered into a collaboration and licensing agreement (the Cooperation Agreement) with Genentech, Inc., a member of the Roche Group, to develop next-generation antibody-drug conjugates (ADCs) targeting several tumor indications based on the company's proprietary Ying'en unique effective payload antibody conjugate (DUPAC) platform.
DUALITYBIO-B (09606) announced that on August 27, 2026, the company entered into a collaboration and licensing agreement (the Collaboration Agreement) with Genentech, Inc. (Genentech), a member of the Roche Group, to develop next-generation antibody-drug conjugates (ADCs) targeting several tumor indications based on the companys proprietary DUPAC platform, which features unique and effective payload antibodies. Under the terms of the Collaboration Agreement, the company will lead the discovery and early global clinical development of the collaboration projects. Genentech will obtain exclusive global rights to the ADCs generated from the collaboration and will assume full responsibility for further clinical development and commercialization after the completion of Phase 1a clinical trials.
According to the Collaboration Agreement, the company will receive a $45 million upfront payment and is eligible to receive potential development, regulatory, and commercialization milestone payments totaling over $1 billion for all collaboration projects. Additionally, the company will be entitled to receive tiered royalties based on the annual net sales of approved products resulting from the collaboration.
The Board of Directors believes that the Collaboration Agreement will enable the company to address the growing unmet medical needs of cancer patients whose tumors have developed resistance or are refractory to the classes of payloads currently relied upon by approved ADCs. The company has established the DUPAC platform (a new family of mechanism-based effective payloads designed to overcome payload resistance) to tackle these challenges and allow each ADC project to broaden its relevance to tumor biology. By combining the companys payload innovations and early global clinical development capabilities with Genentechs deep oncology expertise and global development reach, the collaboration will enable both parties to advance collaboration projects more rapidly and at a larger scale, bringing differentiated medicines to more patients worldwide.
DUPAC is the company's proprietary ADC payload platform featuring a novel mechanism of action and is one of four ADC technology platforms developed by the company, the others being DITAC, DIMAC, and DIBAC. DUPAC encompasses various payloads, including DUP5, DUP9, and DUP10, each of which exerts its effects through unique anti-tumor mechanisms. Preclinical data for the ADCs based on DUP5 and DUP9 (including activity in tumor models relatively insensitive to topoisomerase inhibitor payloads and tolerability in non-human primates) has been presented at multiple medical conferences, including AACR 2025 (Abstract 5454), the AACR-NCI-EORTC International Conference 2025 (Abstracts B129 and B130), and AACR 2026 (Abstract 2657).
