Johnson & Johnson's new prostate cancer drug and head and neck cancer drug amivantamab released positive data at ESMO. JPMorgan said the pipeline has differentiated potential, but the valuation is elevated, and maintained a Neutral rating.
It is noted that the European Society for Medical Oncology (ESMO) Annual Congress has just concluded. This is one of the most important academic conferences in the global oncology field, and the data presented there often directly moves the stock prices and pipeline expectations of related pharmaceutical companies. Johnson & Johnson (JNJ.US) has become one of the most closely watched names in the large-cap pharmaceutical sector. In a research report published on October 25, JPMorgan judged that the early data released by Johnson & Johnson in the two major indications of prostate cancer and head and neck cancer provide potential differentiated competitiveness for its innovative drug pipeline. The core highlight is the prostate cancer candidate drug code-named JNJ-6143, with a technology platform called Halda Riptac. This is an investigational therapy for metastatic castration-resistant prostate cancer (mCRPC), an indication that generally refers to patients who have progressed after treatment with androgen receptor pathway inhibitors and represents an advanced population with unmet clinical needs. The Phase 1 clinical data were quite impressive: enrolled patients were highly advanced, with a median of four prior lines of systemic therapy, and more than 30% had previously received taxane chemotherapy and radioligand therapy. In the 50 mg dose cohort selected to advance to Phase 2, the objective response rate (ORR) reached 75%, and after a median follow-up of 5.6 months, 71% of patients remained on treatment, with efficacy unaffected by genomic characteristics. More importantly, in terms of safety, the incidence of Grade 3 or higher treatment-related adverse events in the 50 mg dose cohort was zero. Analyst Chris Schott believes that these data mean JNJ-6143 may have differentiated advantages in both efficacy and tolerability, and panelists at the session also gave high recognition. Johnson & Johnson will accordingly advance the drug into Phase 3 clinical trials and simultaneously expand into metastatic castration-sensitive prostate cancer and combination therapy directions. The analyst expects that by the 2030s, the large prostate cancer market is expected to bring considerable incremental space to the company. The other main thread is head and neck cancer. The fifth cohort of the OrigAMI-4 trial of amivantamab (Rybrevant) combined with pembrolizumab (Keytruda) and chemotherapy showed that in patients with HPV (human papillomavirus)-unrelated head and neck cancer, the combination regimen achieved an objective response rate of 77% (sample of 22 cases), significantly higher than the approximately 36% response rate of pembrolizumab plus chemotherapy in this indication. Johnson & Johnson has already submitted a marketing application for amivantamab in the second-line head and neck cancer indication, while the first-line Phase 3 trial is advancing in parallel. The report believes that the drug is expected to occupy an important position in the HPV-unrelated head and neck cancer field. Even so, the report remains relatively restrained in its attitude toward Johnson & Johnson's stock. The analyst points out that Johnson & Johnson remains one of the large pharmaceutical companies with relatively clean fundamentals, with its innovative drug business leading at high single-digit growth and its medical technology business also shifting toward high-growth sub-sectors. JPMorgan maintains a "Neutral" rating on Johnson & Johnson with a latest target price of US$285. Measured by estimated earnings per share for 2027, the company's valuation has reached 21x, placing it in a clear premium range. Relative to other names in the same group, the stock's upside is limited, which also explains why the analyst remains cautious on the stock despite optimism about the fundamentals.
